Penelitian ini dilaksanakan karena cara detoksifikasi yang biasa dilakukan masyarakat, yaitu melalui perendaman dan perebusan yang berulang, hanya dapat menghilangkan zat racun sebesar 50%. Bahkan terjadi pengurangan kadar zat gizi yang dikandung singkong. Pemanfaatan singkong pahit sebagai bahan pangan yang aman untuk dikonsumsi terbatas hanya sebagai bahan pembuatan tapioka. Padahal jenis singkong ini mengandung pati yang lebih banyak daripada singkong biasa. Selain itu produksi singkong di Indonesia mencapai peringkat ke-3 setelah Brasil dan Thailand. Pada penelitian ini dilakukan cara detoksifikasi menggunakan mikroorganisme, yang selain mampu menghilangkan zat racun juga dapat meningkatkan protein singkong. Caranya, singkong terlebih dahulu dikeringkan di bawah sinar matahari hingga tercapai kadar air sekitar 10%. Setelah kering, singkong dijadikan tepung. Tepung singkong kemudian ditambah air dalam perbandingan (w/v) 1:1 untuk dipersiapkan sebagai substrat fermentasi yang diaduk hingga terbentuk pasta. Pasta singkong diinokulasi dengan suspensi spora kapang rhyzopus oryzar dalam perbandingan (w/v) 1:10, setelah dihomogenkan kemudian dimasukkan ke dalam cawan petri. Cawan petri, kemudian diinkubasikan pada suhu 28 derajat celcius, hingga tampak pertumbuhan miselium (30 jam). Hasil fermentasi dikeringkan dan ditumbuk hingga berbentuk tepung. Jenis singkong pahit yang digunakan dalam penelitian ini diperoleh dari kebun percobaan Balai Penelitian Bioteknologi Tanaman Pangan dan jenisnya diketahui yaitu jenis singkong Adira II, Adira IV, 46.8 dan 39-1-1, dengan kandungan sianida masing-masing sebesar 15.57 mg, 5.61 mg, 10.02 mg dan 24.67 mg per 100 g. Hasil penelitian menunjukkan bahwa penurunan sianida setelah fermentasi terhadap ke-4 jenis singkong pahit tersebut adalah bervariasi untuk setiap jenis singkong pahit. Singkong pahit Adira IV penurunan sianidanya dapat mencapai 100%, kemudian 87% untuk jenis Adira II dan jenis 46.8 serta 18% untuk jenis 39-1-1. Sedangkan peningkatan kadar protein setelah fermentasi sebesar 2 kali untuk jenis singkong 39-1-1, 0.88 kali untuk singkong Adira II, 0.56 kali untuk singkong Adira IV, dan 0.48 untuk singkong jenis 46.8.
